Re: The magic 500mg threshold :)
I have also found NAD+ before bed helps me sleep better. In particular, I get more deep sleep, according to my oura ring. Dr Matthew Walker shares fascinating info on sleep - how important it is for health, and how to improve it. As important as exercise and diet. Really spurred me to focus on that....

Re: Nuchida Claim
You're right, NAD+ has been proven to enter some cells directly by Connexin43. Also some other routes. https://alivebynature.com/exogenous-nad-enters-hypothalamus-intact-decreases-hunger-and-increases-energy-expenditure/ The enzymes to metabolize it to NMN, NR, NAM and other products like very benef...

Re: Exactly Where is Sinclair on Rolling Back Age?
It is amazing. Reading the book gave me some background I didn't have before that makes it more believable. I used to think Dr Sinclair kind of "lucked" into his role with Resveratrol, somewhat like Dr Brenner did in discovering NR pathway to NAD+. But he actually had a major Eureka breakt...

Re: NAD+ precursor increases aerobic performance.
Thanks, but I'm not sure what you are referring to. They feed the mice 400mg/kg a day of NR for 5 weeks, which would be something like 2000 mg a day for a 70 kg human. Dr Brenners study gave humans 1000 mg a day NR for 3 weeks. Do you mean the different dosage, delivery method, or time length might ...

Re: NAD+ precursor increases aerobic performance.
These were YOUNG mice, which is likely why NR by itself did not help. Would really like to see if NR by itself increases endurance in older mice, like NMN does. Another thing that stands out to me is, NR increased NAD+ levels in the muscle. In the recent study by Dr Brenner , NR did NOT increase NAD...

Podcast by Dr Sinclair and Dr Mark Harmon - Sept 11, 2019
New podcast by Dr Sinclair and Dr Mark Harmon. Mostly rehash but a few interesting things I noticed: 33:30 Sinclair - there is a spike in 2-3 hours, then it goes back down (NAD+ levels from taking NMN) 34:30 Sinclair - Lenny just had a paper that came out yesterday that show parkinsons patient do b...

Re: NAD+ IV increases NAD+ levels in blood plasma 400% in humans
Sinclair has suggested that the microbiome in blood removes a lot of the nicotinamide from NMN and NR before it's taken up by the gut, so maybe some of this is true of NAD+ as well. The exact method is multi-pronged and "messy" according to him but maybe we're getting closer to finding th...

Re: NAD+ IV increases NAD+ levels in blood plasma 400% in humans
Interesting. But how do we know the NAD+ isn't simply metabolized to something else in the first 2 hours? The authors propose 2 possibilities: Either NAD+ and/or the primary metabolites tested are being efficiently sequestered during this first 2–6 h period or secondary metabolites are also being f...
