What exactly does NAD+ do?

Post by Johnnyk »

I've read that NAD is fuel for Sirtuins.
But I was wondering, what exactly does that mean?

First a little background of my understanding.
  1. I understand that the Sirtuin genes produce Sirtuin enzymes.
  2. The Sirtuin enzyme silences the "mating type gene".
  3. When DNA damage occurs, the Sirtuin enzyme goes off to fix the problem.
  4. The mating type gene is now activated because the Sirtuin enzyme has gone
  5. The mating type gene and stops any reproduction activity.
  6. When the Sirtuin enzyme returns after it has done its repairs, it again turns off the mating type gene, allowing reproduction to continue.
So when we take NAD does that mean that
  1. The Sirtuin genes produce more Sirtuin enzymes than they normally would? OR
  2. The existing Sirtuin enzymes are more "energized" and do their job better? OR
  3. Something else?
And finally, does increasing NAD+ help fuel all seven Sirtuins (1-7)?

I think this gives an easy to understand view about what is going on with Sirtuins and the relationship with NAD+
