Can Boosting NAD+ Cause Negative Effects for Those with MTHFR, COMT or MAOA Gene Mutations?

Q1. Just curious if taking NMN, NAD+ or NR can cause negative side effects for those with decreased methylation abilities (MTHFR), degrading catecholamines (slow COMT), or oxidizing serotonin, dopamine, epinephrine, norepinephrine (slow MAO-A). Could it cause heightened anxiety or insomnia? If so, is there a way to counter this?

Q2. Has anyone (regardless of genes) experienced sleep disturbances or anxiety from boosting their NAD+ levels?

I take both NAD+ Sublingual Tablets and NMN Sublingual Tablets as a combination.
The NMN creates disturbing sleep patterns if I take it late in the day where the NAD+ seems to enhance my sleep quality what ever time I take it.. :idea:

Hi James, here are a few key sentences from the interesting article mentioned by Drdavid:
Increased NAD levels enhance glycolysis and fuel cancer cells. In fact, nicotinamide phosphoribosyltransferase (Nampt), a rate-limiting enzyme for NAD synthesis in mammalian cells, is frequently amplified in several cancer cells.
Continuous replenishment of NAD promotes the proliferation and survival of fast-dividing cancer cells because elevated NAD levels enhance glycolysis ..
In other words, cancer cells have a very active metabolism and multiply quickly. As part of this, they depend on high levels of NAD for their metabolism and have found ways to increase their available NAD, for example by increasing Nampt levels compared to normal cells.

The article is talking about the development of different anti-cancer agents which work by trying to decrease NAD levels by targeting the Nampt levels. They summarize the results of experiments using the various agents, but also note that NAD is essential for all normal cells:
Although Nampt inhibitors are promising candidates for preventing tumor cell growth, Nampt is also essential for normal cells. In fact, whole body deletion of Nampt results in embryonic lethality, and muscle-specific Nampt deficient mice exhibit progressive muscle degeneration (57, 97). Moreover, retina-specific Nampt deficient mice exhibit severe vision loss (98). Therefore, it is important to consider these adverse side effects while using Nampt inhibitors in clinical settings.

Thank Dr. Patrick for clarifying this. Unfortunately, I have been traveling for the last couple weeks and missed this post. Your explanation is important for anyone that has been diagnosed with cancer or has had cancer in the past. If there are cancer cells according to this research they are very capable of using NAD to divide. Thus, NAD may not be appropriate as a supplement for these people.
Another interesting fact is the current research on Nampt inhibitors to actually combat cancer. We live in an exciting age in medicine.

I have an MTHFR gene mutation and I haven't noticed any issues thus far. My thinking is clear and my physiology seems to be functioning well. I am curious if any anti aging supplements can affect someone with MTHFR. Can MTHFR affect aging?

Boosting NAD+ may have negative effects for certain gene mutations. For example, people with the MTHFR gene mutation may experience negative side effects from taking NAD+ supplements.
