Why Intranasal Delivery Is Highly Effective
Intranasal delivery is the one of the quickest and most effective ways to deliver NAD⁺ and its precursors, NMN and NR, into the body. The nasal mucosa’s rich blood flow allows for optimal drug absorption while avoiding the first-pass effect. ⑴
The first pass effect refers to drugs getting metabolized at specification locations in the body, most often the liver, which results in the drug’s reduced concentration in systemic circulation or at the target site.

NAD+ Can Cross the Blood-Brain Barrier
As a protective layer surrounding the brain, the blood-brain barrier’s main purpose is to prevent harmful agents from reaching the brain. Research has shown that certain methods can be used to create a passageway through the barrier in order to allow therapeutic agents to reach their target.
In a study published in September of 2018, NAD⁺ was administered through Intraperitoneal Injection and Intravenous injection in mice. The injections demonstrated that NAD⁺ crosses the blood brain barrier to enter the hypothalamus intact, in mice.
“In conclusion, our results demonstrate that exogenous NAD⁺ is effectively imported into the hypothalamus and increases hypothalamic NAD content.” ⑶
Neuroprotection from Intransal NAD+ Delivery
In one study on rats with transient focal ischemia, intranasal administration of NAD⁺ was found to significantly increase NAD⁺ levels in the brain, while being neuroprotective.
