Clinical Trial: Evaluating the Efficacy of Liposomal NMN

A double-blind, placebo-controlled clinical trial was conducted with 15 healthy men aged 40 and older to measure the effects of NMN supplementation. Participants were randomly assigned to one of three groups (5 per group) and took their assigned supplement daily after breakfast for four weeks.

Placebo Group: Maltodextrin (4g/day)
Standard NMN Group: Non-liposomal NMN (350 mg/day)
Liposomal NMN Group: Liposomal NMN (350 mg/day)

Blood NAD+ levels were measured at four time points:

1. Before supplementation (baseline)
2. One hour after the first dose
3. After four weeks of daily supplementation
4. Four weeks after stopping supplementation

This study design allowed researchers to assess not just how effectively each form of NMN raised NAD+ levels, but also how long those effects persisted after supplementation ended.

Liposomal NMN Increased Blood NAD+ Levels By 84%

After four weeks of supplementation, the Liposomal NMN Group exhibited an 84% increase in blood NAD+ levels, rising from 28.6 µM (baseline) to 52.5 µM.

Although NAD+ levels declined four weeks after participants stopped supplementation, they remained elevated at 36 µM compared to the baseline of 28.6 µM.

Encapsulated NMN Boosted NAD+ More Effectively Than Standard NMN

Among all groups, Liposomal NMN produced the greatest increase in NAD+ levels after four weeks, outperforming both Standard NMN and placebo.

Which NAD+ Precursor Is Best?

NAD+

This central coenzyme is directly involved in redox reactions and serves as a substrate for energy production enzyme activity, including sirtuins and poly(ADP-ribose) polymerases (PARPs), which regulate cellular metabolism, DNA repair, and epigenetic modifications.

NMN

This NAD+ precursor, which is directly converted to NAD+ through the NMNAT enzyme in the Salvage Pathway, has been shown to effectively increase intracellular NAD+ levels in numerous clinical trials, thereby enhancing mitochondrial function, energy production, and the activity of NAD+-dependent enzymes.

NR

This NAD+ precursor, which is converted to NAD+ through the Preiss-Handler Pathway, has also demonstrated the ability to increase NAD+ levels in cells. It supports cellular repair mechanisms, particularly those involved in DNA damage response and oxidative stress mitigation. By buttressing NAD+ levels, NR promotes the activation of sirtuins and PARPs, which protect genomic stability and regulating cellular metabolism.

Trigonelline promotes cellular NAD+ production differently than NMN or NR. This methylated form of niacin offers several advantages: exceptional stability in the bloodstream, slow release into target tissues, non-flushing, protection against stomach digestion, and direct NAD+ increase in muscle tissues.

Liposomes Increase Bioavailability

With our Liposomal products, approximately 90% of the payload reaches the bloodstream intact, regardless of what the payload is.

For example, with our Liposomal Green Tea Extract around 90% of the EGCG reaches the bloodstream.

With regular Green Tea extract, around 9% of EGCG reaches the bloodstream.

Hence the study below shows it is about 10x more bioavailable.

Regular Apigenin is around 30% bioavailable, so you can expect our Liposomal Apigenin to be approximately 3x more bioavailable than regular Apigenin.

Although we don't have research on bioavailability of all our Liposomal products, below we show research that demonstrates increased bioavailability of 3 to 40x when using liposomes in some of the most popular supplements we produce.

LIPOSOMAL DELIVERY

What are the benefits of liposomal delivery? Are liposomes really better?

When taken sublingually, approximately 30% of a 125 mg dose gets absorbed under the tongue, with the remainder being swallowed. At higher dosages, the % goes down, as the capillaries are full and have limited transport.

A much greater % makes it to bloodstream with liposomal delivery. Liposomes also protect the payload from degradation once in the bloodstream.

Lastly, Liposomes release the payload over a much longer period – up to 24 hours. This is better when taking larger dosages.

The rapid delivery to the bloodstream with Sublingual can be an advantage when taking before exercise. The energy surge most users feel is also nice confirmation that it is doing something.

What are the liquid and powdered liposome differences in bioavailability and stability?

Our liquid and powdered liposomes have approximately the same availability in the bloodstream, but the powdered liposomes release the payload more slowly over 24 hours. We don’t know if that makes it more effective than a faster release.

Powdered liposomal products have a shelf life of over two years, rather than the 12 months or so for liquid liposomal products when stored at room temp.

What's the difference between Liposomal vs. Delayed & Extended Release Caps?

Delayed Release & Extended Release Caps protect ingredients from stomach acid only. They delay, but don't really sustain ingredient release.

Liposomes protect the payload through the GI tract and into the bloodstream, then release more slowly up to 24 hours (depending on the liposome).

In addition to a big increase in bioavailability, they are truly sustained release. Any supplement known to have poor bioavailability should use liposomal delivery.

Are any of your liposomal products heat-sensitive?

The liposomal powder capsule products are not heat sensitive.

The liposomal gels however should not be exposed to heat exceeding 115 degrees Fahrenheit for more than a few days. Liposomes break down a bit when exposed to extended periods of high heat.
